Growth regulators mediated flavonoid production from callus and cellular suspension of seagrass halodule pinifolia (miki, den hartog)

Seagrasses, a marine angiosperm used in folk medicine as diuretic, anti-cancer, anti-inflammatory, antiseptic and antirepellant. The study was led to look at the impact of growth regulators on callus induction, cellular suspension and flavonoid content from the leaf and rhizomes of Halodule pinifolia. Different types of synthetic auxins (2, 4-D, IAA, NAA, IBA) and cytokinins (6-BAP, kinetin, zeatin) were used in the study with distinct concentrations. Findings revealed the maximum yield of biomass and flavonoid content from the rhizome derived calli (82.19%) of H. pinifolia with the help of 2, 4-D (1.5 mg L-1) and 6-BAP (1 mg L-1). The calli morphology was observed to be yellowish and friable in nature. Flavonoid content (52.11 ± 3.25 g/L DW) was found to be more on 14th day old calli, as they are healthy, rapid cell division and the values obtained are significantly different (P≤0.05). Enhancement of flavonoid production by the precursor or elicitors should be examined using the cellular suspension of seagrasses which will help food and pharmaceutical industries as nutritional supplements or elements for neutraceutical functional foods.
